US: number of religious sisters declines by over 2,000 in one year ...

The number of religious sisters in the United States fell from 57,113 to 55,045 in a one-year period, according to data in the 2011 and 2012 editions of The Official Catholic Directory.

During the same period:

  • the number of priests fell slightly from 40,271 to 40,203
  • the number of seminarians fell from 5,247 to 5,015, with most of the decline occurring among religious-order (and not diocesan) seminarians
  • the number of religious brothers fell from 4,650 to 4,518
  • the number of permanent deacons rose from 17,436 to 17,816

Novel therapeutic advancement in search for heart muscle progenitor cells: New hope for heart attack patients

ScienceDaily (Nov. 22, 2012) ? Breakthrough in heart research: The research team from Professor Katja Schenke-Layland of the Fraunhofer Institute for Interfacial Engineering and Biotechnology IGB in Stuttgart has discovered cell surface markers that enable the identification and isolation of living functional cardiovascular progenitor cells (CPCs). For the first time, therapeutically relevant CPCs can be derived from induced-pluripotent stem cells (iPS) cells. CPCs, which are typically only found in fetal development, can become all of the different cell types of the heart and can integrate into heart muscle tissue after injection.

An estimated 17 million people die from cardiovascular disease each year. Although mortality rates are declining, heart attacks are still among the most frequent causes of death in the developed world. Often, the cause of a heart attack is the closure of a coronary artery that supplies blood to the heart, which kills heart muscle cells. Cardiomyocytes, which are the heart muscle cells responsible for the contraction of the heart, are not able to regenerate after a heart attack. The massive loss of cells and tissue, and the highly restricted regeneration capacity of the adult heart, lead to an impaired blood supply throughout the body that drastically affects a patient's quality of life. To restore the heart's function after a major heart attack, clinicians require functionally mature cardiomyocytes that perform like the native cells in the adult heart to replace the cells that were killed.

The production of such functional cardiomyocytes from well-defined cardiovascular progenitor cells (CPCs) is the focus of the research team led by Prof. Dr. Katja Schenke-Layland from the Fraunhofer Institute for Interfacial Engineering and Biotechnology IGB in Stuttgart and her colleagues, Dr. Ali Nsair of the University of California Los Angeles (UCLA) and Prof. Dr. Robb MacLellan of the University of Washington in Seattle, who have now succeeded in identifying such cells in a mouse model. The work could revolutionize the treatment of heart disease.

Development of heart muscle cells from precursor cells

Myocardial cells -- as well as endothelial cells and smooth muscle cells -- develop from CPCs during the embryonic development of humans and other animals. There has been a significant amount of research effort towards discovering a path for the clinical application of these cells in patients. The reason for the lack of success is that the markers that help to identify CPCs, such as Islet1 or Nkx2.5, are located in the nucleus of the cells. The use of these cell markers modifies the cells rendering them therapeutically unusable, making the identification of safe cell-surface markers essential.

Surface markers identified for cardiovascular progenitor cells

On this task, the research team of Professor Katja Schenke-Layland from the Fraunhofer IGB in Stuttgart, Prof. Dr. Robb MacLellan and Dr. Ali Nsair of the University of California Los Angeles (UCLA), where Schenke-Layland previously worked before returning to Germany to join the Fraunhofer-Gesellschaft's Attract Program, focused their research. With success: They were able to identify two markers, the receptors Flt1 (VEGFR1) and Flt4 (VEGFR3), on the surface of CPCs with which these cells can be clearly identified while fully preserving their biological function. This discovery allows scientists to isolate clinically relevant cardiovascular progenitor cells that can be functionally matured.

In the search for surface markers, the researchers investigated the cardiovascular progenitor cells using microarray gene expression profiling. These studies show exactly which genes are active at a specific point in time. The resulting data from this analysis were compared to the sequencing data from existing databases of already known as cell markers.

From induced-pluripotent stem cells, cardiovascular progenitor cells are developed

Encouraged by the success of being able to identify and isolate living CPCs, the researchers sought out to derive the cells from induced-pluripotent stem (iPS) cells. For this purpose, they used a method for which the Japanese scientist Shinya Yamanaka was recently awarded the 2012 Nobel Prize for Medicine. This work, published just six years ago, demonstrated that only four proteins are responsible for the embryonic state of cells (Takahashi K, Yamanaka S. Cell 2006, 126 (4): 663 -676). He brought those four genes into differentiated -- mature and specialized -- cells, which then returned them back to an embryonic state. From these cells, which he called iPS cells, scientists can develop all cells of the body, such as liver cells, nerve cells or heart muscle cells.

In their study, the researchers used cells from a mouse strain in which the cells are labeled with a visible green fluorescent protein (GFP) that can be identified with a fluorescence microscope. The cells from these mice were then reprogrammed with the same four genes discovered by Yamanka, resulting in iPS cells that could be easily identified.

In a next step, the researchers cultured the GFP-labeled iPS cells in the laboratory under different conditions with cell-influencing solutions such as growth factors. "Using our newly established cell surface markers, we could detect and isolate the Flt1 and Flt4 positive CPCs in culture," says Schenke-Layland. "When we cultured the isolated mouse CPCs then in vitro, they actually developed -- as well as the embryonic stem cell-derived progenitor cells -- into endothelial cells, smooth muscle cells and more interestingly into functional heart muscle cells."

iPS cell-derived CPCs integrate into the living mouse heart

But how do the developed CPCs behave in living organisms? Can these cells really integrate into tissue and regenerate heart muscle? To answer these questions, the scientists injected the GFP-labeled CPCs into the hearts of living mice. After 28 days, the researchers analyzed the hearts and saw that the green fluorescent cells had developed into beating heart muscle cells and had fully integrated into the myocardial tissue of the mouse.

Enormous potential for heart research

Researchers have long tried to stimulate the regeneration of heart muscle cells. For this purpose, they inject stem cells or stem cell-derived cardiomyocytes into the heart. Although the majority of studies found a slight improvement in heart function, in most cases, neither long-term integration nor the differentiation of the cells into heart muscle has been demonstrated.

The result of the group from Schenke-Layland, Nsair and MacLellan provides the first opportunity to generate functioning heart muscle cells, which integrate into the heart muscle. "We are currently focusing on research with human iPS cells. If we can show that cardiovascular progenitor cells can be derived from human iPS cells that have the ability to mature into functional heart muscle, we will have discovered a truly therapeudic solution for heart attack patients," hopes the scientist.

The work of the research group has been funded by the German-American funding from the Federal Ministry of Education and Research (BMBF) and the California Institute for Regenerative Medicine (CIRM), as well as the Fraunhofer-Gesellschaft (Attract Program), the Ministry of Science, Research and the Arts of Baden-W?rttemberg, and the US National Institutes of Health (NIH).

Syria rebels capture army base in eastern oil region

AMMAN (Reuters) - Syrian rebels captured an army base in an eastern oil province on Thursday, further weakening President Bashar al-Assad's control in the strategic region bordering Iraq.

The capture of the artillery base on the outskirts of Mayadeen, a town on the Euphrates river near some of Syria's main oilfields, followed rebel takeovers of military installations in the north and centre of the country this week.

Recent rebel momentum shows the increasing potency of the mainly Sunni Muslim fighters trying to topple Assad, who belongs to the Alawite minority linked to Shi'ite Islam. But insurgents have often had to retreat quickly after making advances to avoid strikes by the president's air force.

"The Mayadeen military base fell at 8.30 a.m. (0630 GMT)," Abu Laila, an official in the Military Revolutionary Council in the province, told Reuters. He said 44 rebel fighters had been killed in the operation to capture the base.

"The whole countryside, from the Iraqi border and along the Euphrates to the city of Deir al-Zor, is now under rebel control," he said.

Another opposition source in contact with rebels confirmed the base, 42 km (26 miles) south-east of Deir al-Zor, had fallen.

Video posted online showed rebels on motorcycles and trucks apparently inside the base waving victory signs as smoke rose from two buildings. Artillery pieces could be seen on the ground and a tank transporter stood abandoned.

Severe restrictions on non-state media make it impossible to verify opposition reports independently.

Activists say 38,000 people have been killed in the 20-month uprising which threatens to draw in regional Sunni and Shi'ite Muslim powers. Hundreds of thousands have fled the country and 2.5 million are displaced, aid groups say.

Western states, anxious to avoid another costly Middle East conflict and wary of backing rebels who include Islamist militants, have stayed on the sidelines, although France and Britain formally recognized a newly formed opposition coalition as the sole representative of the Syrians this month.

Russia, which along with China has blocked three resolutions which could have led to U.N. sanctions against Assad, criticized on Thursday proposals for NATO to deploy Patriot missiles in Turkey near the Syrian border.

"This would not foster stability in the region," Foreign Ministry spokesman Alexander Lukashevich said.

REBEL GAINS

The capture of the Mayadeen base leaves Assad in control of just three major army bases in Deir al-Zor province, said Sheikh Nawaf al-Bashir, a local tribal leader. He said rebels now held the main road to Iraq, from the outskirts of Deir al-Zor city to the border crossing of Albu Kamal.

The rebel move follows the capture last week of a military airport on the Iraqi border southeast of Mayadeen. In the last five days rebels have also stormed a special forces base near Aleppo, Syria's commercial hub, and an air defense position in the southern suburbs of Damascus.

In another setback for Assad, his forces pulled back on Thursday from three positions south of the town of Maarat al-Numan, on the highway linking Damascus to Aleppo, according to the opposition Syrian Observatory for Human Rights.

The area has seen heavy fighting between rebel forces, which have held the town for several weeks, and soldiers camped in a nearby military base, just 500 meters from the highway.

Near Damascus, opposition campaigners said fighting continued around the south-western suburb of Daraya and the army kept up heavy bombardment of the town, where Free Syrian Army fighters appeared to be entrenched.

To the east, clashes were also reported in the Damascus neighborhood of Jobar, which is adjacent to the main Abbasid Square. A mother, her daughter and her sister's husband were killed in shelling aimed at pushing back rebels, activists said.

Rebels in the south Damascus district of Hajar al-Aswad showed video of a captured air defense officer they identified as Colonel Bashir al-Saleh, flanked by two masked rebels carrying AK47s.

So far Assad's core military units, composed mainly of members of his Alawite minority sect, have prevented a sustained rebel push into the heart of the capital itself. The rebels have yet to hold a major Syrian city.

But activists say the rebels are gaining strength in Damascus, partly because they are being joined by fighters from outlying regions. While Assad's forces control main road junctions there have been guerrilla attacks in the last few days near Damascus Airport and rebels have expanded control of the mixed urban and farmland regions around the capital.

The Observatory said army shells struck a building next to Aleppo's Dar al-Shifaa hospital on Wednesday, one of the main rebel medical centers, killing 15 people. Most of the dead were fighters but a doctor and three children also died, it said.

Labor Unions Launch Six-Figure Ad Blitz Opposing Spending Cuts To Medicare, Medicaid, Education

One running theme of deficit reduction negotiations is that both parties support domestic spending cuts in their opening bids -- Republicans demand them, and Democrats champion them alongside tax increases for high income earners.

Now a coalition of three labor unions -- AFSCME, SEIU and National Education Association -- are launching a six-figure ad buy pressuring swing-state Senate Democrats and targeted House Republicans to oppose spending cuts to Medicare, Medicaid and education -- three items that neither side has taken off the table in talks about defusing a looming austerity bomb.

The ad campaign reflects an effort to upend a Washington status quo where Democrats are pushing for spending cuts to lure Republicans into supporting tax increases. The groups are nervous that President Obama and Democrats may go along with a deal that cuts the safety net.

"We haven't heard any assurances [from the White House or Democratic leadership] on anything at this point," Mary Kusler, director of government relations for NEA, told reporters on a conference call Tuesday. "That's why we're continuing to remind everybody that we need to craft a deal that puts middle class families first."

On the call, the three groups unveiled a new survey they commissioned, conducted by the Mellman Group, which said voters would oppose a "grand bargain" that goes after programs like Social Security, Medicare, education and funding for local police and firefighters.

"None of these programs need to be cut to reduce the deficit," said AFSCME's director of federal government affairs Charles Loveless. "The overwhelming majority, 53 percent, want taxes increased on the wealthy, and want to see these core programs protected."

Three 30-second TV ads titled Jobs Not Cuts airing in Colorado, Virginia and Missouri call on Democratic Sens. Michael Bennett (CO), Mark Udall (CO), Mark Warner (VA), Jim Webb (VA) and Claire McCaskill (MO) to tame the deficit "not by cutting programs that families rely on most" and work toward protecting Medicare, Medicaid and education from cuts."

Peter Colavito, SEIU's director of government relations, said "these leading Democrats are important voices within their Senate caucus" in brokering a deal.

Separate 1-minute radio ads called Can't Wait seek to jolt members of the Republican-controlled House into extending the middle income tax cuts, which GOP leaders are refusing to do unless Democrats agree to also continue the lower top-bracket tax rates.

"There's one thing that both parties can agree on: we should raise taxes on the middle class. But if Congress fails to act soon, that's exactly what will happen," says a narrator in the ad, which targets Republican Reps. Pat Meehan (PA), Mike Fitzpatrick (PA), Don Young (AK) and Jo Ann Emerson (MO) in their respective states.

"Congress should act quickly to preserve the middle class tax cuts," the narrator says, "and that will take leaders willing to put people over partisan politics."

Approximately 4 million animals are euthanized at shelters every year, and this is a conservative estimate. NASA: Astronauts could survive Mars radiation

Radiation on the surface of the Red Planet is roughly similar to that experienced by astronauts on the international space station, report NASA scientists. The findings help researchers understand the planet's capacity for supporting life and will assist NASA in preparations for future manned flights to the planet.?

The rover's initial radiation measurements ? the first ever taken on the surface of another planet ? may buoy the hopes of human explorers who may one day?put boots on Mars, for they add more support to the notion that astronauts can indeed function on the Red Planet for limited stretches of time.

"Absolutely, astronauts can live in this environment," Don Hassler, of the Southwest Research Institute in Boulder, Colo., told reporters during a news conference today (Nov. 15).

Hassler is principal investigator of Curiosity's Radiation Assessment Detector instrument, or RAD. RAD aims to characterize the Martian radiation environment, both to help scientists assess the planet's past and current potential to host life and to aid future manned exploration of the Red Planet. Since Curiosity landed on?Mars?in August, RAD has measured radiation levels broadly comparable to those experienced by crewmembers of the International Space Station, Hassler said. Radiation at the Martian surface is about half as high as the levels Curiosity experienced during its nine-month cruise through deep space, he added.

The findings demonstrate that?Mars' atmosphere, though just 1 percent as thick as that of Earth, does provide a significant amount of shielding from dangerous, fast-moving cosmic particles. (Mars lacks a magnetic field, which gives our planet another layer of protection.)

The $2.5 billion?Curiosity rover?is getting a bead on the nature of this shielding. RAD has observed that radiation levels rise and fall by 3 to 5 percent over the course of each day, coincident with the daily thickening and thinning of the Martian atmosphere, researchers said.

Hassler stressed that RAD's findings are preliminary, as Curiosity is just three months into a planned two-year prime mission. He and his team have not yet put hard numbers on the Martian radiation levels, though they plan to do so soon.

Sotheby's to sell Mick Jagger's love letters

LONDON (AP) ? Handwritten letters from Rolling Stones frontman Mick Jagger to his former lover Marsha Hunt will be auctioned in London next month.

Hunt is an American-born singer who was the inspiration for the Stones' 1971 hit "Brown Sugar" and bore Jagger's first child.

Sotheby's says she has tasked the auction house with selling 10 letters written from the set of Jagger's film "Ned Kelly," which was shooting in Australia.

Sotheby's says the "passionate and articulate" letters sent in the summer of 1969 show a "poetic and self-aware" 25-year-old Jagger.

The auction house said Saturday that the collection, which includes song lyrics and a Rolling Stones playlist, is expected to fetch between 70,000 and 100,000 pounds ($111,300 and $159,000) and will go under the hammer on December 12.

What does the "cancel for work reasons" benefit cover? | Travel ...

Due to the sometimes uncertain conditions of vacation time with an employer, many travelers are interested in the ?cancel for work reasons? benefit that is often included in travel insurance policies. If previously granted vacation time is revoked and a traveler is forced to cancel ?a vacation, this benefit can be very valuable.

In order to claim this benefit, employees will be required to obtain a written statement from a company officer or human resources manager demonstrating revocation of previously approved time off. There must be proof that time off for the scheduled vacation existed and was then overturned.

If an individual is self-employed, they may still claim this benefit, but they will be required to submit proof of self-employment and proof of the requirement to work.

Self-employment can be substantiated by tax records, incorporation documents, and the like. In order to prove revocation of planned time off, a self-employed traveler may submit a notarized personal statement that explains the reasons for the revocation.
